In addition to what Rei_12 said, if your post gets more than 5 downvotes (I think its 5, at least), your post is hidden with the message, "This post has been hidden due to low karma -- click to show."

Also downvoting doesn't cause you to lose karma, you can only gain karma.

But what about the Karma that's shown on your dashboard?

The karma that's shown on your dashboard is the karma you gain from your posts.
