The name of the "real deal" program is, actually, AutoTune. It's made by a software company called Antares. The I AM T-Pain app is actually powered by Antares software, which makes it the cheapest way to get AutoTune (if you already have an iphone / ipod touch, that is).

Pretty sure theres no stand-alone Antares AutoTune software, so you'd need something like Garage Band, Ableton Live, Cubase… something which will load VST or AU plugins.
As far as I know, some freewares like Audacity can run VST plug-ins. I've never tried an Autotune with it myself so I can't confirm anyway.

I can confirm that AutoTune works w. Audacity as a VST and AU plug.
