Super Smash Brothers - Sorry Popo... Nana's moved on

Wierdly enough, they never actually say that they're related. Even the official word of Nintendo for the the relationship of these two is "up to the players imagination". It's just that them being siblings is the most common theory and people simply took it as fact after a while.
