It's the improv stuff that always seems to be the most legendary. For example in Star Wars: Episode V we had:

"I know."

Apparently that scene had a bazillion takes and at that point, Harrison Ford was just so fucking done with doing it over and over. Turns out his frustration made was brilliantly in character.
