New thread notification

If you use Gmail, a desktop client, or any decent web app for email, this shouldn't be a problem. Threaded messages and filters make it simple to filter all your notifications into a separate folder (or label, if you're using Gmail).
In Gmail, go to Settings>Labels>Create New Label, then type "KnowYourMeme" (or whatever you want, really), click Create. Now go to Filters and click the "Create a new filter" link at the bottom of the page. Where it says "From," type "noreply@knowyourmeme.com" and click the Test Search button to confirm that it's going to apply the labels correctly. Now click the Next Step button. Tick the following: "Skip the Inbox," "Apply the Label: (Select the label you created earlier)," and "Also apply filter to X conversations below." Once that's done, click Create Filter and you're done.
In Yahoo! Mail, navigate to Options>More Options>Filters>Add New Filter. Give it a name, then in the Sender field, enter "noreply@knowyourmeme.com" and leave the drop-down menu to "contains." Then where it says "Move to Folder," select "[New Folder]." Click the Save Changes button, then when it prompts you to name the folder, well, name the folder. After that, you should be good to go.

If you don't want to receive any notifications, or simply want to receive notifications from only a select few threads, unsubscribe from the threads you don't want notifications from from the subscription management page.
