In fact, the game gives Mario a sidequest to reach Floor 50 after completing the entire story, so the intended point at which the player is expected to obtain the Strange Sack is in the postgame. Regardless, with enough practice, preparation, knowledge of combat tactics and a high skill with blocking/Superguarding enemy attacks, an expert player would be able to reach deeper floors of the Pit of 100 Trials as early as it is unlocked, ie. in the middle of Chapter 1, and get the Strange Sack very early.
